苯妥英钠、苯巴比妥、扑痫酮、卡马西平和丙戊酸类药物治疗癫痫大发作均有较好疗效。治疗癫痫的药物如：苯妥英钠因治疗剂量和中毒剂量接近，为防不测临床已少用。苯巴比妥由于镇静作用明显影响注意力、记忆力，且容易出现兴奋、多动和攻击行为，学龄儿童中常不选用。

苯妥英钠治疗癫痫：本品为抗癫癎药、抗心律失常药。治疗剂量不引起镇静催眠作用，动物实验证明，本品对超强电休克、惊厥的强直相有选择性对抗作用，而对阵挛相无效或反而加剧，故其对癫癎大发作有良效，而对失神性发作无效。

卡马西平治疗癫痫：能起到稳定作用，能降低神经细胞膜对Na+和Ca2+的通透性，从而降低细胞的兴奋性，延长不应期;也可能增强GABA的突触传递功能。抗惊厥的机制尚不清楚，类似苯妥英，对突触部位的强直后期强化的抑制，限制致痫灶异常放电的扩散。

苯巴比妥治疗癫痫：用作镇静和催眠药物，适用于治疗神经过度兴奋引起的失眠症，能引起安稳的睡眠。但不宜长期服用，因其会引起全身无力、呕吐、头痛等副作用。可由二乙基丙二酸酯与尿素在乙醇钠存在下经缩合反应制得。
